Suas capacidades gráficas são impressionantes graças à Unreal Engine 3, assim como os sons e a jogabilidade. Tudo começa com Dominic Santiago, mais conhecido como Dom, resgatando Marcus Fenix de uma prisão para que ambos possam ajudar a conter a invasão de uma raça alienígena, os Locust. A partir de então, o jogador encarna o soldado Fenix, e parte para um dos combates mais sanguinários vistos na história do videogame. Durante sua jornada, Marcus vai enfrentar monstros horríveis e gigantescos, devendo sempre se esconder do poderoso fogo inimigo, evitando morrer prematuramente.
